Factors responsible for pig infections caused by the significant respiratory pathogens porcine circovirus type 2 (PCV2), porcine reproductive and respiratory syndrome virus (PPRSv), and Mycoplasma hyopneumoniae (M. hyopneumoniae) were investigated in a cross-sectional study. Gastrointestinal (GI) parasites, along with hyo and Actinobacillus pleuropneumoniae (App), pose a considerable health risk in Uganda. Data on infection management procedures were collected via a structured questionnaire. The sampling process included 90 farms and 259 pigs. Four pathogens were detected in sera samples using commercially available ELISA tests. The Baerman's method was used to characterize parasite species found in faecal samples. Through the use of logistic regression, an assessment was made to identify the risk factors for infections. The results of the study revealed an individual animal seroprevalence of PCV2 at 69% (confidence interval 37-111). A seroprevalence of 138% (95% confidence interval 88-196) was observed for PRRSv, 64% (95% confidence interval 35-105) for M. hyo, and an exceptionally high 304% (95% confidence interval 248-365) for App. In terms of prevalence, Ascaris spp. was 127% (95% confidence interval 86-168), Strongyles spp. was 162% (95% confidence interval 117-207), and Eimeria spp. demonstrated a very high prevalence of 564% (95% confidence interval 503-624). Up until now, there have been no efforts to cultivate this intracellular bacterium from the filarioid host in vitro. As a result, the current study employed a co-culture system of embryonic Drosophila S2 cells and LD cell lines for the purpose of cultivating Wolbachia from Dirofilaria immitis microfilariae (mfs) harvested from affected dogs. 1500 microfilariae (mfs) were inoculated into shell vials, which were subsequently supplemented with Schneider medium, and employed both cell lines for the procedure. Beginning at day zero, the initial introduction of the bacterium, along with observations of its multiplication and establishment, continued with each medium change from day 14 until day 115. By utilizing quantitative real-time PCR (qPCR), 50-liter aliquots from each time point were assessed. When averaging the Ct values from the different parameters (LD/S2 cell lines and mfs with/without treatment), the S2 cell line not subjected to mechanical disruption of mfs displayed the most numerous Wolbachia cells, as measured by qPCR. Fluorescent microscopy and viability staining will be employed in further experiments to determine the level of Wolbachia infection and cell viability in the cell line.
